高效液相色谱法测定瑞香狼毒根、茎、叶、花中7,8-二羟基香豆素的含量

摘　　要：建立了反相高效液相色谱法测定瑞香狼毒根、茎、叶、花中7,8二羟基香豆素含量的方法,同时利用二极管阵列检测器建立了7,8二羟基香豆素标准品纯度检测法和植物提取样品中7,8二羟基香豆素紫外光谱鉴定方法。色谱柱为AlltechC18柱(250mm×4.6mm,5μm);流动相为甲醇∶水(40∶60,V/V);流速为1.0mL/min;二级管阵列检测器检测,检测波长326nm;柱温28℃。线性范围1.4～880mg/L;线性回归系数大于0.9999;检出限为0.2mg/L(3σ)。测得瑞香狼毒根、茎、叶、花中7,8二羟基香豆素百分含量分别为0.37%、0.58%、1.76%和160%;其中叶样品中7,8二羟基香豆素的回收率为95.8%～100.8%;RSD为1.92%(n=6)。该方法简便,准确,线性范围宽,重现性好,适于对含有该成分的药物和药材进行质量评价。A determination method of 7,8-dihydroxy-coumarin in roots, stems, leaves and flowers of Stellera chamaejasme L. by reversed-phase high performance liquid chromatography was established and at the same time the purity of 7,8-dihydroxy-coumarin was detected by means of Diode-Array Detector. The chromatographic column, Alltech C-18 (250 mm x 4.6 mm, 5 mum), methanol-water (40:60, V/V) mobile phase with 1.0 mL/min flow rate, the detected wavelength (326 nm), and the column temperature (28degreesC) were adopted. The calibration curve was linear under the concentration of 1.4 similar to 880 mg/L, the correlation coefficient was over 0.9999, the detected limit was 0.2 mg/L(3sigma). The contents of 7,8-dihydroxy-coumarin in roots, stems, leaves and flowers of Stellera chamaejasme L. were 0.37%, 0.58%, 1.76% and 1.60%, respectively. The recovery was 95.8% similar to 100.8% and RSD was 1.9% (n = 6). The method is simple, accurate and has wider linear range. The method can be adapted to evaluate the quality of medicinal plants that contain 7,8-dihydroxy-coumarin.
