摘  要:目的 探讨不同类型的应激反应内分泌 免疫变化的特点及其与H2基因多态性的关系。方法 制备束缚应激(倾向于精神性应激和慢性应激 )和热应激 (倾向于躯体性应激和急性应激 )实验动物模型。采用碳粉廓清试验测定小鼠单核吞噬细胞系统的功能;通过放射免疫技术测定血清中皮质醇的水平。同时采用PCR技术检测MudoEb5和MudoEb7为H2 Eb位点遗传多态性标志,分析应激免疫和遗传的关系。结果 束缚组较对照组的吞噬指数有明显的下降(6. 54vs7. 89,P<0. 05),但血清皮质醇水平无统计学差异(9. 37vs7. 99,P>0. 05 );热应激组较对照组的吞噬指数有明显的下降 ( 6. 82vs7. 89, P<0. 05),血清皮质醇有显著性升高(17. 53vs7. 97,P<0. 05)。束缚组MudoEb5+与MudoEb5 的吞噬指数无显著性差异(6. 23vs6. 63,P>0. 05),MudoEb7+与MudoEb7 的吞噬指数有显著性差异(5. 85vs7. 39,P<0. 05);热应激组MudoEb5+与MudoEb5 的吞噬指数无显著性差异 (7. 29vs7. 36, P>0. 05),MudoEb7+与MudoEb7 的吞噬指数有显著性差异(7. 00vs8. 38,P<0. 05)。结论 在不同类型的应激条件下,小鼠的内分泌 免疫变化有所不同,其免疫变化与H2 Eb多态性有关。ObjectiveTo investigate the mechanism about endocrine-immune change in different types of stress and the association about those changes with H2 gene polymorphism. MethodsAnimal models, restraint stress and heat stress, were established. The innate immune response was detected by charcoal clearance assay and the content of cortisol in the serum was measured by radioimmunoassay. MudoEb5 and MudoEb7 were determined with PCR for H2-Eb allelic polymorphism. ResultsThere is a obvious decline in Charcoal Clearance Index for the restraint stress group compared with the control group(6.54 vs 7.89,P<0.05); there then is not a obvious increase in cortisol for the restraint stress group compared with the control group(9.37 vs 7.97,P>0.05);there is a significant decline in Charcoal Clearance Index for the heat stress group compared with the control group(6.82 vs 7.89,P<0.05); there also is a significant increase in cortisol for the heat stress group compared with the control group(17.53 vs 7.97,P<0.05); there is little relation between the change of immune in stress and MudoEb5 as well as close relation between that change and MudoEb7. ConclusionDifferent types of stress have strong suppressive effects on the immune system, and the degree is linked with H2 genetic polymorphism. There may be different mechanism about endocrine-immune change in different types of stress.
