鳞翅目昆虫染色体的研究方法

摘　　要：本研究以鳞翅目昆虫睾丸为材料，在常规压片法基础上引进空气干燥法进行改良，得到高清晰度的昆虫染色体图谱。睾丸在不同的低渗液里处理后，置入卡诺氏液固定，然后用空气干燥法制片，Ｇｉｅｍｓａ液染色。结果表明这一方法制备所得到的昆虫染色体图，不仅染色体分散好，而且染色体的细微结构较清晰。On the basis of air dry-squash,the improved methods for obtaining good cytological preparla-tions from testicular tissues of Lepidoptera are deseribed.Testes were pretreated with various hypotonic solu-tions and fixed in Carnoy's. Slides were prepared by cell suspension-air dry technues. Air-dried slides werestained directly with Giemsa's stain. Well-spread chromosomes with good morphological characteristics werefound in spermatogonial cells and spermatocytes of all stages.A detailed characterization of the mitotic chromosome complement and lst meiotic division in males of Agrotix ypsilon, A. segetum, Mytimna separata and Chilo suppressalis were shown.
