摘  要:利用国内几个黄瓜品种(系)“津研四号”、“长春密刺”、“农大秋棚一号”、“农大秋光”的成熟胚为外植体,在MS+2,4-D 1.0mg·L^(-1)+KT 0.5 mg·L^(-1)培养基上诱导愈伤组织,在MS+IAA 0.1mg·L^(-1)+KT 1.0mg·L^(-1)培养基上形成胚性愈伤组织。基因型差异明显,“农大秋棚一号”胚性愈伤组织的诱导率最高,可达60.2%。胚性愈伤组织在MS+IAA 0.1mg·L^(-1)+KT 1.0mg·L^(-1)+ABA 1.0mg·L^(-1)+5%蔗糖培养基上,可长期继代增殖并保持胚性。利用液体继代培养基,用胚性愈伤组织建立起分散良好的细胞悬浮系,悬浮细胞的细胞质浓。胚状体发生能力强,悬浮细胞转入到固体继代培养基上可迅速生长,并在1/2 MSO+1%蔗糖培养基上胚状体萌发成苗。Somatic embryogenesis was gained from embryos of Chinese cucumber cultivars'Jinyan No. 4','Changchunmici', 'Nongdaqiupeng No. 1' and'Nongdaqiuguang'. It had best results first on basal MS medium contains 2, 4-D 1. 0 mg·L^(-1) and KT 1. 0 mg·L^(-1) for callus inducing, then on basal MS meduim containing IAA 0. 1 mg·L^(-1) and KT 1. 0 mg·L^(-1) for embryoid formation. There were significant difference in the frequency of embryogenesis amony 4 cultivars in which the highest one was'Nongdaqiupeng No. 1'(about 60. 2%). The basal MS medium containing IAA 1. 0 mg·L^(-1), KT 1. 0 mg·L^(-1), ABA 1. 0 mg·L^(-1) and sucrose 5% was fit for embryogenic callus to be subcultured and retained the ability of embryogensis. The suspension cultures of embryogenic callus were grown by liquid shake culture in the subcultured medium. Suspension cultures were composed of a population of cells that were densely cytoplasmic and potentially embryogenic. Embryogenic suspension cells were transfered on solid subcultured medium, which grew fast and germinated into plantlets on basal 1/2 MS containing sucrose 1%.
