摘 要:利用9个微卫星标记,采用PCR扩增,体积分数8%聚丙烯酰胺凝胶电泳,对96只优质细毛羊基因组DNA的遗传多样性进行了检测。结果表明:除G54279外,8个微卫星化点均具有高度多态性;群体的平均多态信息含量(PIC)为0.768 8,遗传杂合度(h)为0.774 2,有效等位基因数(Ne)为4.7.均高于国内外部分研究结果。证明微卫星可以作为有效的遗传标记用于优质细毛羊的遗传多样性分析。To reveal the genetic diversity of High Quality Merino Sheep,the genetic polymorphism of 9 microsatellites in High Quality Merino Sheep were studied by means of PCR ,polyacrylamide gel (8%). Number of alleles,average effective number of alleles (Ne) of sheep breeds were counted. According to al lele frequencies of nine microsatellites, polymorphism information content (PIC),mean heterozygosity (h) were calculated for sheep breeds. The alleles were studied by using UVI-08 software. The results showed that 8 of 9 microsatellites loci were high polymorphic, but G54279 were medium polymorphic,so the 8 of microsatellites loci were effective markers for the ananlysis of genetic polymorphism among High Quality Merino Sheep. The average PIC (0. 768 8) ,h (0. 774 2) ,Ne (4. 7) of the population were higher than that of other sheep breeds which were reported in the document, genetic diversity were plenty.it can be studied and analysised deeply.
