摘  要:以多年生黑麦草的成熟种子为外植体,应用正交设计方法,对愈伤组织的诱导和分化培养基进行了优化筛选。结果表明,愈伤组织诱导的最佳因素组合:2,4-D(7mg/L)+6-BA(0.1mg/L)+NAA(0.3mg/L)+CH(0.1g/L)(特拉华);2,4-D(7mg/L)+6-BA(0)(尤文图斯、托亚)。特拉华愈伤组织分化的最佳因素组合:6-BA(0.1mg/L)+NAA(0.8mg/L)+ZT(0.1mg/L)+Cu2+(8.96mg/L);6-BA为0.2mg/L时,托亚的愈伤组织分化率最高。比较三个品种的出愈率和分化率:特拉华最高,托亚次之,尤文图斯最低。Mature seeds of three perennial ryegrass (Lolium perenne) varieties were cultured for callus induction and differentiation on MS basic medium containing different concentrations of 2,4-D, 6-BA , NAA and so on. The results showed that there were significant influences of different concentrations of the supplements on callus induction and differentiation. For Delaware, 7mg/L 2,4-D,0.1mg/L 6-BA, 0.3mg/L NAA and 0.1g/L CH is propitious to callus induction. For Juventus and Taya callus induction, 7mg/L 2,4-D and 0 mg/L 6-BA is more favorable. As for callus differentiation,the best ingredients and concentrations for Delaware are 0.1mg/L 6-BA, 0.8mg/L NAA, 0.1mg/L ZT and 8.96mg/L Cu^(2+). (0.2mg/L) 6-BA is propitious to Taya callus differentiation.In general, Delaware proved to be the best of the three, Taya comes second and Juventus the worst.
