摘  要: 目的:观察长期使用外源性一氧化氮(NO)抑制剂Nω-硝基-L-精氨酸(L-NNA)和促进剂谷氨酸单氨钠(MSG)后高血压大鼠心室壁厚度和心肌血管紧张素Ⅱ(ANGⅡ)质量分数的变化。方法:双肾双夹法建立高血压大鼠模型,分别用NO抑制剂和促进剂腹腔注射进行干预,用测微尺测量大鼠心室壁的厚度,放免法测定心肌ANGⅡ的质量分数。结果:(1)各组大鼠心室壁厚度的变化:高血压组大鼠30、70d较对照组增厚(P<0 05);注射L-NNA组左室壁增厚明显,在30、70d较对照组和高血压组差异明显(P<0 05);而注射MSG组左室壁较注射L-NNA组薄,但仍厚于对照组(P<0 05)。(2)心肌ANGⅡ的质量分数:高血压组大鼠心肌ANGⅡ的质量分数逐渐升高,均较对照组有显著差异(P<0 05);注射L-NNA后,ANGⅡ的质量分数升高明显,其中30、70d与高血压组比较差异显著(P<0 05),而注射MSG组ANGⅡ的质量分数较注射L-NNA组明显降低(P<0 05),但仍然高于对照组(P<0 05)。结论:NO生成增多可减少心肌局部ANGⅡ的合成和分泌,对高血压所导致的心肌肥厚有一定的抑制作用。Aim: To investigate the influence of ectogenic nitric oxide inhibitor and accelerator on the thickness of cardiac ventricle and ANG Ⅱ in hypertensive rats. Methods: After having gotten the hypertensive rats by clamping bilateral kidney arteries, we intervened the hypertensive rats by ectogenic nitric oxide inhibitor and accelerator and measured the thickness of cardiac ventricle of hypertensive rats with micrometer and the concentration of ANG Ⅱ with radioimmunity. Results: (1) Changes of the thickness of cardiac ventricle: the thickness of left ventricle had no significant changes at 7 d in hypertensive rats, but accompanying elongation of hypertension, the thickness of left ventricle was increased markedly at 30 d and 70 d(P<0.05). After the rats had been treated with L-NNA, the left ventricle was thicker than the control or hypertensive rats at 30 and 70 d(P<0.05). The results also showed that MSG could reduce the thickness of left ventricle at 70 d, yet, this changes did not back to control or hypertensive rats (P<0.05). (2)Changes of ANG Ⅱ in myocardium: in hypertensive rats, the concentration of ANG Ⅱ gradually increased and was higher than the control rats (P<0.05). After the rats had been treated with L-NNA, the concentration of ANG Ⅱ increased significantly at 30 d and 70 d (P<0.05). When (given) MSG, the hypertensive rats got a little ANG Ⅱ, however, still more than the control group (P<0.05).Conclusion:Nitric oxide may reduce the synthesis and secretion of local angiotensin Ⅱ and (inhibite) myocardium to be thickening resulted from hypertension.
