摘  要:目的比较两种不同溶解体系提取健康汉族成年男子骨髓蛋白质组双向电泳 (2 -di mensionalelectrophoresis ,2 -DE)图谱的差异。方法人骨髓分别用A、B两种不同的溶解体系提取蛋白质 ,以固相pH梯度 (IPG)等电聚焦为第一向 ,SDS -PAGE垂直电泳为第二向进行 2 -DE。采用ImageMaster 2DElite(Version 3.0 1)软件分析 2 -DE图谱。结果两种溶解体系提取的蛋白质样品分别获得 10 2 8± 4 2个蛋白质点和 130 0± 5 8个蛋白质点 ,其中 871个蛋白质点在两张图谱中可互相匹配 ,其余 15 7和 4 2 9个蛋白质点分别为两种溶解体系各自特有。用两种溶解体系可获得 14 5 7个蛋白质点。Objective To compare the 2-dimensional electrophoresis (2-DE) maps of human bone marrow proteome extracted by two different solution systems. Methods Healthy adult man bone marrow proteome of Han nationality was resuspended in two different solution systems. The extracted protein was separated by 2-DE, immobilized pH gradient (IPG) isoelectric focusing as the first dimension, then vertical SDS-PAGE as the second dimension. The maps were analyzed by Image Master 2D Elite (Version 3.01). Result There were 1 028±37 and 1 300±58 protein spots in each maps, of which 871 spots were matched in the two maps. The rest 157 and 429 protein spots were not matched. Combianation of the two solution systems would obtain 1 457 protein spots. Conclusion The satisfactory 2-DE may be achieved by combining the two different solution systems.
