摘  要:目的研究5种氟喹诺酮类药物对大肠埃希菌耐药突变株的选择能力,比较其防耐药变异能力。方法肉汤法富集1010CFU/ml大肠埃希菌(ATCC25922)和25株临床分离菌,采用平板稀释法测定莫西沙星、加替沙星、帕珠沙星、左氧沙星和环丙沙星对大肠埃希菌的MIC99、暂定防变异浓度(MPCpr)及MPC。结果莫西沙星、加替沙星、左氧沙星、帕珠沙星、环丙沙星对大肠埃希菌(ATCC25922)的MPC分别为0072、0048、009、009、006μg/ml。细菌耐药选择指数(MPC/MIC99)分别为4、68、69、9、12。环丙沙星、加替沙星、帕珠沙星、莫西沙星对25株大肠埃希菌临床分离菌MPCpr90值分别为2、1、2、1μg/ml,选择指数(MPCpr90/MIC90)分别为16、4、16、2。结论莫西沙星和加替沙星限制大肠埃希菌耐药突变菌株的选择能力强于环丙沙星和帕珠沙星。OBJECTIVE To measure mutant prevention concentrations(MPC) in vitro of fluoroquinolones for Escherichia coli and compare the capacity of fluoroquinolones to prevent the resistant mutant strain. METHODS The E. coli strain ATCC25922 and 25 clinical isolates were enriched in broth,and the bacterial concentrations were adjusted to 10 10 colony forming units per milliliter. The minimal inhibitory concentration(MIC), MIC for 99% of input cells (MIC 99 ), provisional MPC(MPCpr) and MPC of moxifloxacin, gatifloxacin, pazufloxacin, levofloxacin and ciprofloxacin for E. coli were determined by agar plates dilution method. RESULTS The MPC of moxifloxacin,gatifloxacin, levofloxacin, pazufloxacin and ciprofloxacin for E. coli strain ATCC25922 were 0 072, 0 048, 0 09, 0 09 and 0 06μg/ml, and the MPC/MIC 99 were 4, 6 8, 6 9, 9 and 12, respectively. The MPC for 90% of the isolates(MPCpr 90 ) of ciprofloxacin, gatifloxacin, pazufloxacin and moxifloxacin for 25 E. coli clinical isolates were 2, 1, 2 and 1μg/ml,and the MPC pr90 /MIC 90 were 16, 4, 16 and 2, respectively. CONCLUSIONS The capacity of moxifloxacin and gatifloxacin for restricting the selection of E. coli resistant mutants is stronger than that of pazufloxacin, levofloxacin and ciprofloxacin.
