摘 要:目的探讨复方黄连抑制鼻咽癌细胞裸鼠移植瘤生长的分子机制。方法应用高通量互补DNA(cDNA)表达芯片杂交以及逆转录 — 聚合酶链式反应技术 ,分析复方黄连处理后CNE1和HNE1鼻咽癌细胞裸鼠移植瘤组织NF—κB活化相关基因信使RNA(mRNA)的表达。结果复方黄连在抑制移植瘤生长的同时 ,尚可改变包括TRAF5、TRAF6、TRAP2、p67、p68激酶、IκBα以及LAP—1在内的一些NF—κB活化相关基因在鼻咽癌移植瘤组织的表达。其中 ,IκBα和TRAF6基因的表达为上调 ,其余基因的表达均为下调。结论复方黄连对鼻咽癌细胞裸鼠移植瘤生长的抑制作用 。Objective To investigate the molecular mechanism of Compound Coptidis Formula (CCF) responsible for the growth-inhibiting of transplanting tumor tissues with nasopharyneal carcinoma (NPC) cells. Methods The procedures of hybridization with cDNA microarray and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect mRNA transcription of NF-κB-activating related genes in the transplanting tumor tissues of NPC cell lines CNE1 and HNE1, following the treatment with CCF.Results It was shown that the growth of transplanting tumor was significantly inhibited among the experimental animals following the use of CCF. Meanwhile, the mRNA transcription activity of NF-κB-activating related genes, like TRAF5, TRAF6, TRAP2, p67, p68 kinase, IκBα and LAP-1, were obviously changed as well, showing up-regulated transcription activities of IκBα and TRAF6 and down-regulated transcription activities of the remainders. Conclusion It is suggested from this study that CCF brings about growth-inhibiting effect on the transplanting NPC tumor via the regulating mechanisms on the expressive activity of NF-κB gene in the transplanting tumor tissues.
