糖尿病大鼠肾脏蛋白聚糖Ⅱ和转化生长因子-β_1mRNA的动态变化

摘  要:目的了解糖尿病大鼠肾皮质中蛋白聚糖Ⅱ(PGⅡ)mRNA和转化生长因子-β1(TGF-β1)mRNA随时间的变化及相互关系。方法用链脲佐菌素(STZ)造成大鼠糖尿病模型,在造模成功后第3、7、14、30、60、90天分别用免疫组化法检测糖尿病组和对照组肾脏Ⅳ型胶原分布,半定量RT-PCR法检测肾皮质PGⅡmRNA和TGF-β1mRNA的含量。结果与对照组相比,糖尿病组肾脏Ⅳ型胶原含量增加(除第3、7天,其余时点均P<0.05),TGF-β1mRNA从第7天起升高(P<0.05),PGⅡmRNA后期升高(第60天P<0.05鸦第90天P=0.01)。PGⅡmRNA和TGF-β1mRNA呈正相关关系(r=0.7熏P<0.01)。结论STZ诱导的糖尿病大鼠肾皮质TGF-β1mRNA和PGⅡmRNA先后升高,TGF-β1mRNA可能是引起PGⅡmRNA升高的原因之一。Objective To investigate the dynamic changes of proteoglycan Ⅱ ,PGⅡ, and transforming growth factor-β1,TGF-β1, mRNA in renal cortex of streptozotocin ,STZ,-induced diabetic rats. Methods Diabetes were induced in male SD rats by single intraperitoneal injection of 1.0% STZ ,70mg/Kg,. At the 3rd 7th 14th 30th 60th and 90th days after diabetic model established, PGⅡ and TGF-β1 mRNA values in renal cortex were determined by RT-PCR and collagen Ⅳ values were measured by immunohistochemistry. Results Comparing to normal rats, diabetic rats had higher levels of collagen Ⅳ,P<0.05), at all time points except the 3rd and 7th day, . TGF-β1 values gradually increased ,P<0.05), except the 3rd day,. PGⅡ values increased from the 60th day, at the 60th day, P<0.05)?鸦 at the 90th day, P=0.01,. TGF-β1 and PGⅡ had a significant positive correlation,r=0.7, P<0.001,. Conclusion PGⅡ and TGF-β1 mRNA increased significantly in renal cortex of diabetic rats. TGF-β1 may be one of the stimulating factors for the expression of PGⅡ.
