PRELIMINARY STUDY OF CRYOPRESERVATIVE TECHNIQUE FORVIABLE HEART VALVES AND BLOOD VESSELS

摘  要:用改良的方法建立了活性同种瓣膜和血管库,观察了用三抗培养液37℃消毒24h和低温保护剂(CPM)在室温条件下对组织的影响。结果表明,冻存复温的组织培养24h后,糖利用率和谷-草转氨酶(AST)明显降低,72h后乳酸脱氢酶(LDH)则明显升高,此后与新鲜组织趋于一致;三抗培养液消毒和CPM对组织均无明显影响;组织学观察发现冻存复温组织生长较新鲜组织稍晚。用该技术低温保存的活性同种瓣膜和血管进行10例临床移植,获得满意的效果。提示改良法低温冻贮活性生物瓣膜和血管技术已初步成功。A bank of viable heart valves and vessels was established by modified methods. The effects of three-antibiotic-culture medium used for sterilization at 37℃ for 24h and of cryopreservative medium (CPM) at room temperature on tissues were observed. The results indicated that the usage of glucose and the level of AST in the freeze-thawed tis- sues declined obviously after 24h of culture and LDH increased significantly after 72h. Three-antibiotic-culture medium sterilization and CPM had no effects on tissues. The freeze-thawed tissues grew slightly later than the fresh tissues. Satisfactory clinical re- sults have been obtained in 10 cases of homotransplantation using the viable heart valves and vessels preserved by the present technics, which suggests that this modified method is pretty usable.
