摘 要:目的 观察大豆苷元对心肌细胞损伤的影响并探讨其可能机理。方法 用体外细胞培养方法 ,培养乳鼠心肌细胞 ,制备过氧化氢损伤模型 ,测定细胞存活率、上清液中乳酸脱氢酶活性 (LDH)、肌酸激酶 (CK)、丙二醛 (MDA)、超氧化物歧化酶 (SOD)、一氧化氮 (NO)和一氧化氮合酶 (NOS)含量。结果 大豆苷元 (10 - 16 0 μmol/L)各剂量都能显著地提高过氧化氢损伤心肌细胞的存活率和用MTT法测得的A5 70nm值 ;显著地抑制过氧化氢损伤引起心肌细胞的LDH和CK的释放 ;显著地减少过氧化氢损伤的心肌细胞MDA的生成 ,提高过氧化氢损伤的心肌细胞SOD的活性 ;显著地抑制过氧化氢损伤的心肌细胞NOS活性 ,降低NO水平。结论 大豆苷元对过氧化氢损伤的心肌细胞具有保护作用。Objective To observe the protective effects of daidzein on the myocardial cells injured by hydrogen peroxide (H 2O 2) and to study its action mechanisms with the method of cell culture.Methods Myocardial cells from 2-3 days rats were cultured in the 1640 medium and the cellular injury was induced by H 2O 2.the viability of myocardial cells, dehydrogenase(LDH), creatine kinase(CK), malonaldehyde (MDA), surperoxide dismutase (SOD ), nitric oxide (NO) and nitroxide synthase(NOS) in the medium were measured.Results Daidzein(10-160μmol/L)significantly increased the viability of myocardial cells and A570nm of MTT , inhibited release of dehydrogenase(LDH) and creatine kinase(CK), decreased the content of malonaldehyde (MDA) and increased surperoxide dismutase ( SOD ) activity,decreased the contents of nitric oxide (NO) and nitroxide synthase(NOS) of myocardial cells injured by H 2O 2.Conclusion Daidzein has protective effects on myocardial cells injury and the effects are beneficial to prevention and treatment of Myocardial reperfusion injury(MRI).
