胃康宁对胃癌细胞血管内皮生长因子(VEGF)及其受体表达的影响

摘　　要：目的:研究胃康宁对胃癌细胞血管内皮生长因子(VEGF)及其受体Flt、KDR表达的调控作用。方法:以不同剂量的中药复方胃康宁制剂给SD大鼠灌胃制备含药血清,然后以含药血清培养胃癌细胞,共同培养两代后,分别用免疫组化法和RT-PCR检测不同剂量组胃癌细胞中VEGF及其受体Flt、KDR的表达情况。结果:胃康宁高、中、低剂量组对P53、VEGF和Flt的免疫组化检测与对照组比较,差异有非常显著性意义(P<0.01);中、低剂量组与高剂量组比较,差异有非常显著性意义(P<0.01)。胃康宁高、中、低剂量组对VEGF及其受体Flt、KDRmRNA表达检测与对照组比较,差异有非常显著性意义(P<0.01);中、低剂量组与高剂量组比较,差异有非常显著性意义(P<0.01),提示与用药剂量有一定的关系。结论:胃康宁对胃癌细胞血管内皮生长因子及其受体Flt、KDR的表达有抑制作用。Objective: To observe the influence of modified Zhoufei Tang (MZT) on life quality in the stationary phase of chronic obstructive pulmonary disease (COPD). Methods: 60 cases of COPD were classified to Ⅰ - ⅡA grades and allocated to 2 groups randomly. The control group was not treated with any medicines and the treatment group was treated by MZT (composed of Radix Codonopsis , Gecko, Ramulus Cinnamomi, Radix Asteris, Flos Far farae, Fructus Schisandrae, Semen Armeniacae Amarum, Semen Per sicae, Lignum Aquilariae Resinatum, Semen Juglandis and Pulmo Caprinus) . The influence of life quality in both groups were observed after 6 months. Results: after treatment, the total scoring (TMS) of life quality, daily life ability scoring (F1S), social activity scoring (F2S) anxiety psychologic scoring (F4S) in treatment group showed no difference with those before treatment (P > 0. 05), indicating that the life quality was maintained in the original stable level, while the depression psychologic scoring (F3S) was decreased in comparison with that before treatment ( P < 0. 05). In control group, TMS, F1S, F3S were increased after treatment (P < 0. 05), indicating that the life quality is decreased. But TMS, and F4S F1S, F3S and F4S were significantly different before and after treatment in both groups (P < 0. 05, P < 0. 001). It is indicated that the decreasing tendency of life quality is postponed in treatment group. Conclusion: MZT can stabilize the life quality in the stationary phase of COPD.
