摘 要:以聚乙二醇(PEG)1000为相转移催化剂,负载型杂多酸为催化剂,在乙酸介质中,用双氧水氧化3,4,5-三甲氧基甲苯制备了合成辅酶Q10的重要中间体2,3-二甲氧基-5-甲基-1,4-苯醌,也称辅酶Q0.研究获得了较合适的反应条件:反应温度在30~35℃,24g 3,4,5-三甲氧基甲苯原料需要0.4g PEG-1000催化剂,乙酸和50%双氧水各40mL,20mL负载型杂多酸催化剂.反应液经过水洗、萃取和结晶提纯后,辅酶Q0纯度达到97%,收率为90%.2,3-Dimethoxy-5-methyl-1,4-benzoquinone called coenzyme Q0, a key fragment in coenzyme Qn series and a key intermediate for synthesizing coenzyme Q10 was prepared from 3,4,5-trimethoxy toluene using polyethylene glycol (PEG-1000) as phase transfer catalyst (PTC), as well as supported heteropolyacid as acid catalyst, acetic acid as medium, hydrogen peroxide as oxidant. The suitable condition was found that 40 mL acetic acid, 40 mL 50% hydrogen peroxide, 20 mL supported heteropolyacid catalyst and 0.4 g PEG-1000 are required for 24 g raw material 3,4,5-trimethoxy-toluene and the reacting temperature is 30-35°C. Via washing reaction medium with water, extraction with dichloromethane and removal of solvent, the crystal of product was prepared with the purity and yield of 97% and 90% respectively.
