摘 要:沙枣茎段在MS+NAA(0.2mg/L,单位下同)+ZEA(1)培养基上首先诱导形成愈伤组织,然后转接到MS+ZEA(0.2)+BA(2)培养基上经4次继代产生大量不定芽。沙枣的茎段也可以扦插在MS+BA(0.5)或MS+NAA(0.1)+BA(0.5)培养基上直接获得大量不定芽。以上两种途径获得的不定芽均可以在MS_0培养基上生根而培育成小苗,而通过后一种途径获得的不定芽具有很好的遗传稳定性。为进一步做遗传转化及耐盐筛选工作提供材料。The callus was produced from the stem segments of Elaeagnus angustifolia on the MS medium containing NAAO(2mg/L) and ZEA (1 mg/L). A lot of leaf buds were produced after the callus was transferred 4 times on the MS medium containing ZEA (0. 2 mg/L) and BA (2mg/L). Many leaf buds were directly produced by inserting the stem segment of Elaeagnus angustifolia in the MS medium containing BA(0. 5 mg/L) or containing NAA(0. 1 mg/L) and BA (0. 5 mg/L). The leaf buds produced by the two ways stated above can root in MS in about 15 days and produce plants later. The leaf buds produced directly from the stem segments possess a good genetic stability which can be applied further in the transformation and screening of the salt tolerant plants.
