摘  要:目的探讨红花黄素(SY)预处理对心肌缺氧/再给氧损伤保护作用的机制。方法制备大鼠心肌细胞缺氧模型,以酚妥拉明、卡托普利分别处理20min、5h后,给予缺氧预处理或SY预处理,以缺氧3h/再给氧1h期间心肌细胞的生存率、搏动频率、培养液中LDH、MDA和SOD释放为观测指标。结果酚妥拉明处理细胞20min后,给予缺氧预处理或SY预处理,其细胞搏动频率、细胞生存率均下降;培养液中LDH、MDA释放增多,SOD活性下降,与I/R组情况相近;卡托普利处理组细胞搏动频率、细胞生存率增加,LDH、MDA释放减少,而SOD活性增加,与单纯缺氧/再给氧组比较有明显差异。结论α受体阻断剂酚妥拉明可取消缺氧预处理和SY预处理对心肌细胞的保护作用,提示SY预处理保护作用的机制可能与α-肾上腺素受体和CPK介导有关。Objective:To understand the intracellular mechanism of preconditioning protection by safflor yellow.Methods:Investigated the effects of angiotensin converting enzyme inhibitor(ACEI) and α-adrenoceptor antagonists on the spontaneous beating,the survival tate and release of LDH,MDA and the activity of SOD in cultured redreonatal rat cardio myocytes treated by IP and safflor yellow during the reoxygenation 1h following hypoxia 3h.Results:Preconditioning by hypoxian and safflor yellow increased the spontaneous beating and the survival rate,and decreased the released of LDH and MDA with the rise of SOD.Inhibition of ACEI by captopril and α-adrenoceptor antagonistsby phentolamine abolished the protection by hypoxia preconditioning and safflor yellow with reduction of the beating,Survival rate,activity of SOD,and increase of the release of LDH and MDA.Conclusion:The results indicated that activation of signal transducion path way from α-adrenoceptor antagonists seem to be involved in the cardio protection of precnditioning by safflor yellow.
