摘 要:目的 探讨鲑鱼鱼白DNA(SalmonmiltDNA ,SMD)对小鼠胸腺增龄性萎缩作用的分子机理。方法 10月龄雌性Balb c小鼠按体质量随机分为实验组和对照组 ,在常规饮食的基础上各组每日分别灌胃 333.33和 0mg kg的SMD ,5周后 ,无菌取胸腺 ;应用RT PCR和Northernblot技术检测Genebank登录号为Aw2 0 910 2、U2 3789、X80 2 32、NM 0 0 96 78和AF0 8392 9的基因在对照组和实验组胸腺组织中的表达情况。结果 Aw2 0 910 2、U2 3789、X80 2 32在实验组表达上调 ;NM 0 0 96 78、AF0 8392 9在实验组表达下调。结论 SMD可通过促进细胞增殖相关基因和发育、分化相关基因表达 。Objective To investigate the molecular mechanism of salmon milt DNA (SMD) in age-related involution of mouse thymus. Methods Ten-month-old female Balb/c mice were divided randomly into experiment and control groups according to their weights. The mice in the experiment group were supplied with SMD (333.33 mg/kg) but the mice in the control group were not supplied with SMD. RT-PCR and Northern blot were used to detect the expressions of Aw209102, U23789, X80232, NM009678, and AF083929 genes in all their thymuses. Results The expression levels of Aw209102, U23789, and X80232 genes were higher in experiment group those in control group, but the expression levels of NM009678 and AF083929 genes were lower in experiment group than in control group. Conclusion SMD may reverse the age-related involutions in mouse thymus via up-regulating the expression of proliferation related genes as well as the expression of development and differentiation related genes, and via down-regulating the expression of apoptosis related genes simultaneously.
