摘 要:目的 研究缺氧新生豚鼠心肌缺血再灌注时 β1肾上腺能受体 (β1 AR)的变化及其与心功能的关系 ,以及St.ThomasHospitalcardioplegicsolutionⅡ (ThomasⅡ号液 )和冷血心停搏液对 β1 AR和心功能的影响。方法 建立新生豚鼠常压缺氧模型和离体心脏左心做功模型 ,运用多导生物信号分析系统测定心功能指标 ,采用放射配基结合法测定心肌 β1 AR的密度和亲和力。结果 缺氧新生豚鼠心肌 2 0℃缺血再灌注后心肌 β1 AR的密度显著降低 (P <0 0 1)。ThomasⅡ号液使其降低更为显著 ,而冷血心停搏液则可促进 β1 AR密度的恢复。ThomasⅡ号液对缺氧新生豚鼠心脏缺血再灌注后的心功能无保护作用 ,而冷血心停搏液具有较好的保护作用。β1 AR的亲和力无明显变化 (P >0 0 5 )。结论 缺氧新生豚鼠心肌缺血再灌注后心功能的下降与 β1 AR密度的降低有关 ,与其亲和力关系不明显。ThomasⅡ号液和冷血心停搏液对缺氧新生豚鼠心肌缺血再灌注损伤的不同影响可能与它们对 β1 AR密度变化的作用不同有关。Objective To study the relationship between β 1-aderenergic receptor (β 1-AR) and cardiac function in hypoxic newborn guinea-pig myocardia undergoing ischemic reperfusion, and the effects of St. Thomas Hospital cardioplegic solution Ⅱ (STS) and cold blood cardioplegia (CBC) on the β 1-AR and cardiac function. Methods Normobaric hypoxic guenia-pig model and isolated perfused working heart preparation were established. Cardiac functions were recorded by a multichannel biologic signal analyzer. Myocardial β 1-AR density (B max ) and affinity (K d) were investigated by radial ligand binding. Results Significant decrease of B max was found in hypoxic newborn guinea-pig myocardia suffering from hypothermic (20 ℃)-ishemic reperfusion (P<0.01). More pronounced fall of B max was produced by STS, while significant recovery of B max was improved by CBC. CBC but not STS could effectively protect cardiac function of hypoxic newborn guinea-pig subjected to myocardial ischemic reperfusion. No change of K d was observed (P>0.05). Conclusion The impairment of cardiac function in hypoxic newborn guinea-pig undergoing myocardial ischemic reperfusion might be associated with the decline of B max , but not with K d. Different effects of STS and CBC on cardiac function may result from their different influences on B max .
