摘 要:目的 测定培养大鼠乳鼠心肌细胞内游离钙离子浓度 ([Ca2 + ]i) ,观察KCl、异丙肾上腺素 (Iso)对心肌细胞 [Ca2 + ]i的影响 ,初步探讨其作用机制。方法 采用Fura - 2作为细胞内钙离子的荧光探针负载培养的心肌细胞 ,结合阳离子测定系统检测心肌细胞 [Ca2 + ]i。结果 静息时 ,心肌细胞 [Ca2 + ]i为 83 3±11 6 7nmol/L ,30mmol/L、 5 0mmol/L、 70mmol/LKCl使心肌细胞 [Ca2 + ]i升高 ,且呈剂量依赖性 ;同时10 μmol/L异丙肾上腺素使心肌细胞 [Ca2 + ]i升高。结论 Fura - 2 /AM结合阳离子测定系统可以精确的反映细胞内游离钙离子浓度 ,KCl、异丙肾上腺素升高心肌细胞 [Ca2 + ]i的作用机制不同。Objective To measure free Ca^(2+) concentration of cultured cardiac cells in neonatal rats ,and observe the effect of KCl,isoprenaline(Iso)in [Ca^(2+)]i , and approach initially their mechanism of action. Methods Rat neonatal cardiac myocytes were cultured, Fura-2/AM was loaded as calcium probe.The changes of cyosolic Ca^(2+) were observed by cation meaturement system. Results The value of Ca^(2+) was (83.3±11.67)nmol/L(±s) for 6 quiescent cardiac cells, KCl 30,50,and 70mmol/L triggered a sustained rise in [Ca^(2+)]i in concentration-dependent manner,the same isoprenaline 10mmol/L also produced a sustained elevation in [Ca^(2+)]i. Conclusions The system can accurately reflect cytosolic concentration of free Ca^(2+) ,KCl,Iso can increase [Ca^(2+)]i by different actiong mechanism.
