摘  要:目的 :研究阴道上皮细胞对白色念珠菌生长的抑制作用及雌激素对此作用的影响。方法 :用酶消化法分别分离雌激素处理组、动情间期组和去势组小鼠阴道上皮细胞作为效应细胞 ,白色念珠菌芽生孢子作为靶细胞 ,将效靶细胞共同孵育 9h ,测定白色念珠菌生长抑制率 ,观察其生长密度及透射电镜下超微结构的改变。结果 :白色念珠菌与阴道上皮细胞共同孵育后 ,生长受到抑制 ,生长密度降低 ,超微结构表现为与阴道上皮细胞靠近的念珠菌细胞壁崩解 ,细胞膜连续性破坏 ,甚至细胞器肿胀溶解。雌激素处理组与去势组和动情间期组比较 ,阴道上皮细胞对念珠菌的生长抑制率降低 (P <0 .0 5 )。结论 :小鼠阴道上皮细胞具有天然的抗白色念珠菌作用 ,雌激素能降低此抑制作用。Objective:To investigate the growth inhibition to Candida albicans mediated by vaginal epithelial cells and determine if estrogen affected the acti vity. Methods:C57BL/6J mice vaginae from estrogen treatmen t group, diestrus gr oup and ovariectomized group were excised respectively. The vaginae were dissoci ated into single cell suspension by dispase and collagenaseⅠ. Immunohistochemic al analyse of the separated cells demonstrated that over 80% were vaginal epithe lial cells. The epithelial enriched cells were used as effector cells. Stationar y-phase blastoconidia of C. albicans strains were us ed as target cells . After 9h coincubation of effector(E) with target(T) at various ratios,C. albi cans growth density was observed by upside-down microscopy. The incorpora ted 3 H- glucose was measured by liquid scintillation and the percent growth inhibitio n of C. albicans was calculated. The ultra-structural changes were ob served by tra nsmission electron microscopy.Results:After 9h coincubation o f effector cells with target cells, growth density of C. albicans was visibly reduced and it′s gr owth activity was inhibited. Compared to ovariectomized group and diestrus group , epithelial cells from estrogen-treated mice had less ability to inhibit the g r owth of C. albicans(P<0.05). C. albicans incubated alone showed intact and legi ble ultrastructure while coincubated Candida showed obvio us changes: the cell wa ll ruptured, intact cytoplasmic membrane was damaged, intracellar component d is solved, organellae swollen and even dissolved.Conclusion:Mice vaginal epithelial cells have innate anti-Candida activity i n vitro and the activity was inhibited by estrogen.
