摘  要:目的研究细胞周期蛋白依赖性激酶抑制因子p57Kip2和p27Kip1基因mRNA在胃癌组织中的表达及意义。方法采用RTPCR技术,分别检测p57Kip2和p27Kip1基因mRNA在108例胃癌组织及60例癌旁组织中的表达。结果p57Kip2基因mRNA在胃癌组织中阳性表达率为28.70%,显著低于癌旁组织中的阳性表达率56.67%(P<0.05),p57Kip2基因mRNA相对含量检测胃癌组织为0.59±0.17,显著低于癌旁组织1.63±0.21(P<0.05);p27Kip1基因mRNA在胃癌组织中阳性表达率为23.15%,低于癌旁组织中的阳性表达率31.67%(P>0.05),p27Kip1基因mRNA相对含量检测胃癌组织为1.53±0.23,稍高于癌旁组织1.46±0.35(P>0.05)。结论胃癌组织中p57Kip2和p27Kip1基因蛋白表达的降低与转录机制有关,并在胃癌的发生中存在不同的抑癌途径或不同的调控机制。Objective: To explore the expression of cell cycle protein-dependent anti-kinase gene p57 Kip2 and p27 Kip1 in stomach carcinoma.Methods:Reverse transcription-polymerase chain reaction (RT-PCR) was used to explore the expression of p57 Kip2 and p27 Kip1 in 108 cases of stomach carcinoma and 60 of peri-tumor tissue. Results: The positive rate of p57 Kip2 in stomach carcinoma was 28.70%, which was evidently lower than that in peri-tumor tissue 56.67% (P< 0.05). The comparative content of p57 Kip2 mRNA in stomach carcinoma was 0.59±0.17, which was much lower than the 1.63±0.21 in peri-tumor tissues (P<0.05). The positive rate of p27 Kip1 in stomach carcinoma was 23.15%, which was lower than the positive rate 31.67% (P>0.05) in peri-tumor tissue. The comparative content of p27 Kip1 mRNA in stomach carcinoma was 1.53±0.23, which was slightly higher than that in peri-tumor tissues (1.46±0.35,P>0.05). Conclusion:The differences in the positive rate and comparative content of p57 Kip2 between stomach carcinoma and peri-tumor tissues were significant. If only positive rate and comparative content of p27 Kip1 were concerned, there’s no difference between stomach carcinoma and peri-tumor tissues. The decreased exprssion of p57 Kip2 and p27 Kip1 in stomach carcinoma may be related to the transcription mechanisms, and there’re different anti-tumor pathes and adjust manners.
