摘 要:目的 :比较人肝癌组织与正常肝组织TAL活性差异 ,探讨TAL活性变化在肿瘤组织增殖中的作用 .方法 :取人肝癌组织 2 0例 ,人正常肝组织 10例 ,癌旁组织 10例分别测转醛醇酶活性 ,RT PCR检测转醛醇酶mRNA表达 ,Westernblot检测转醛醇酶蛋白表达水平 .结果 :①转醛醇酶活性在人肝癌组织中 [(6 74± 4 4 )nkat/g]明显高于正常肝组织 [(2 87± 4 4 )nkat/g]及癌旁组织 [(40 1± 2 9)nkat/g],组间存在显著性差异(P <0 .0 5 ) ,正常肝组织与癌旁组织中转醛醇酶活性无显著性差异 .②人肝癌组织与正常肝组织、癌旁组织中转醛醇酶蛋白、核酸表达水平无显著性差异 .结论 :人肝癌组织中转醛醇酶活性较正常肝组织显著升高 ,表现出其在癌变组织中的特异性 .同时提示其活性的改变与翻译后修饰有关 .这为深入研究肝癌的发生机制及癌细胞增殖机制提供了必要的理论依据 .AIM: To compare the activity of transaldolase (TAL) in hepatoma and normal liver tissues and to investigate the role of TAL in the formation of hepatoma. METHODS: Total protein was extracted from the liver tissue of 20 patients with hepatoma and 10 healthy controls, and the activity of TAL was evaluated. The TAL expressions at mRNA level were determined by RT-PCR and those at protein level were determined by Western blot. RESULTS: The activity of TAL in hepatoma tissue [(674±44)nkat/g] was significantly higher than that in normal liver tissues [(287±44) nkat/g] and pericancerous tissues [(401±29) nkat/g] (P< 0.05). The expression of TAL mRNA and protein was not significantly different between hepatoma tissues and normal liver tissues. CONCLUSION: The activity of TAL in hepatoma was significantly higher than that in normal liver tissue, which may be closely related to the neoplastic abnormalities. The change of activity may be associated with post-translational modification.
