摘 要:目的 观察以氨哮素为代表的外界因素对失神经支配红、白肌肉内感觉神经生长因子 (nerve growthfactor,NGF)含量及感觉神经再生的影响。 方法 选用 Wister大白鼠 6 4只 ,体重 2 0 0~ 2 5 0 g,雌雄不拘。平均分成8组 ,每组 8只 (双侧 ,共 16例标本 )。其中 4组为实验组 ,另 4组为对照组 ,分别于神经损伤后 1、3、7和 14 d取材。剪断大白鼠双侧坐骨神经制成腓肠肌 (gastrocnemius,GAS)和比目鱼肌 (soleus,SOL)失神经支配模型。实验组术后每日按2 0 0 μg/ kg剂量给大鼠服用氨哮素 ;对照组术后每日给生理盐水。在各时间点用免疫组织化学方法检测实验组和对照组动物 GAS和 SOL 内 NGF含量 ,另用鸡胚背根神经节培养方法测定各组肌肉提取液感觉神经营养活性。 结果 与对照组相比 ,实验组 SOL 内 NGF含量在术后第 1、3和 7天时增高比较明显 (P<0 .0 1) ;GAS中只有术后第 1天 NGF含量增高明显 (P<0 .0 5 ) ,第 3、7和 14天时实验组 NGF含量与对照组相近 (P>0 .0 5 )。实验组内比较 ,GAS组术后第 1天NGF含量最高 (P<0 .0 1) ,以后 NGF含量逐渐下降 ;SOL 组 NGF含量各时间段相近 (P>0 .0 5 )。实验组两种肌肉间比较 ,在伤后第 7天时 ,SOL内 NGF的含量高于 GAS(P<0 .0 5 )。在对鸡胚背根神经节的神经营养活性?Objective To investigate the influence of clenbuterol on the expression of nerve growth factor (NGF) in denervated red and white muscles and the neurotrophism of the denervated muscles. Methods Sixty four Wister rats, weighed 200 250 g, were divided into 8 groups(8 rats per group), including 4 experimental groups and 4 control groups. The denervated model was made in rats by dissection of sciatic nerves. Clenbuterol was given at a dose of 200 μg/kg per day in the experimental group, saline in the control group. The expression of NGF was measured with immunohistochemistry after 1, 3, 7 and 14 days of injury. The culture methods of dorsal root ganglions of the chick embryos were used to measure the neurotrophism of extracts of the muscles. Results Compared with the control groups, the NGF content of gastrocnemious(GAS) increased on the 1st day (?P?<0.05) and the NGF content of soleus(SOL) increased greatly on the 1st, 3rd and 7th day after injury in the experimental groups (?P?<0.01). In the experimental groups, the NGF amount of GAS reached the highest value on the 1st day after injury(?P?<0.01) and then decreased gradually. And the NGF amount of SOL had slight difference between different time. The NGF content of the SOL was higher than that of GAS on the 7th day (?P?<0.05). The sensory neurotrophism of the extracts was similar between SOL and GAS. Conclusion Clenbuterol can change the expression of NGF in denervated muscles, but the change was different in SOL and GAS.
