摘　　要：用ＨＢｓＡｇ制备了一株分泌乙型肝炎病毒前Ｓ２抗原单克隆抗体（ＭＡｂ）的杂交瘤细胞株（１Ｅ２），小鼠腹水中ＭＡｂ效价达１０＾７。１Ｅ２ＭＡｂ有很强的抑制多聚人血清白蛋白（ｐＨＳＡ）和ＨＢｓＡｇ中前Ｓ２抗原结合的能力；与重组痘苗病毒表达的中分子蛋白（含Ｓ和前Ｓ２抗原）而不与小分子蛋白（含Ｓ抗原）反应；与人工合成的前Ｓ２肽段（Ｎ端１２０－１４６氨基酸序列）特异性结合。A hybrid cell line producing MAb, which could recognize an epitope encoded by the Pre-S(2) region of HBV genome was obtained by hybridoma technique. The MAb-1E2MAb reacted with Pre-S(2) epitope on middle protein of the viral envelope The specificity of 1E2MAb waa confirmed by its capability of binding to the synthetic pep-tide corresponding to N-terminal 120-146 ammo acid sequence of Pre-S(2) region. lE2MAb was used to develop a sensitive enzymoimmunoassay for determination of Pre-3(2) im the sera of HBV infectious patients. The results showed that the titers of Pre-S(2) and HBsAg in HBsAg positive sera were well correlated (r = 0.87, P< 0.01), especially in HBeAg positive cases (r = 0.94, P<0.01).
