体外培养中少突胶质细胞在立体网架上的迁移

摘　　要：应用聚酯纤维在培养基上形成的立体网架,接种生后7天大鼠视神经,然后对少突胶质细胞在三维空间的迁移进行体外培养观察。以抗髓鞘碱性蛋白抗体标记少突胶质细胞,并用扫描电镜和透射电镜对在聚酯纤维上迁移的少突胶质细胞超微结构进行了研究。结果表明,聚酯纤维能引导少突胶质细胞迁移;在其表面,少突胶质细胞的突起相互交错并将其包绕;有些突起已在其表面融合,形成“类膜”结构,有些部位可见有2～3层这样的膜性结构。免疫细胞化学染色结果表明,少突胶质细胞突起在聚酯纤维表面形成的“类膜”结构为髓鞘碱性蛋白阳性。以上结果提示,在体外培养条件下,聚酯纤维能引导少突胶质细胞迁移,在无神经元存在时,少突胶质细胞仍可形成髓鞘碱性蛋白阳性的膜性结构。本文对中枢神经系统髓鞘形成模式进行了讨论。In order to understand the migrating behaviour of oligodendrocytes on the threedimension frame, the polyester fibers (PF) were used on collagen coated coverslips to form a stereoframe on which a small piece of P-7 Wistar rat optic nerve was explanted. The oligodendrocytes were marked with anti-MBP antibody. The ultrastrueture of the oligodendrocytes migrating on the PF was observed under SEM and TEM. The results showed that the oligodendroeytes migrated along the PF and a lot of vesicles were observed around the oligodendrocytes. There were 'analogous membrane structures' (AMS) surounding the PF and two or three layers of the 'AMS' can be observed in some place. The immunocytochemical results revealed that the vesicles around the oligodendrocytes and the 'AMS' surrounding the PF were MBP positive. Thus we believe that the PF are able to induce the migration of the oligodendrocytes in vitro and the oligodendrocytes express MBP in absence of neurons, The models of CNS myelination were discussed.
