摘　　要：本文通过聚合酶链式反应(PCR)方法从鲤鱼脑垂体中分离其总RNA,经过反转录成为第一条链cDNA,再经过PCR扩增,合成了鲤鱼生长激素基因,并把其克隆到大肠杆菌表达载体(pBluescriptⅡKS^+/-)上,获得了重组质粒,又经过PCR证明获得的重组质粒中含有鲤鱼的生长激素基因。Growth hormone genes of fish have a significant growth-promoting effect There have been reported successes of transgenic fish. Here is a research report on cloning growth hormone gene for carp (Cyprinus carpio) .Total RNA is isolated from carp pituitary gland. Then the first strand cDNA is synthesized using oligo (dT)12-18 as primer, total RNA as template and AMV reverse transcriptase. Next the polymerase chain reaction(.PCR) is performed using the first strand cDNA as template, Synthetic 29 oligonucleotides with BamHl and Sall restriction sit as primer and Taq DNA polymerase(94℃60s, 55℃ 30s, 72℃ 50s, 35 cycles). After this, PCR products is cloned into an E. coli expression vector(pBluescriptIIKS+/-). It is verfied that recombinant vector contains 620bp carp growth hormone gene by PCR amplification.Therefore we have gotton carp growth hormone gene.
