摘 要:目的:探讨活细胞免疫荧光法在免疫偶合物制备过程中的应用价值。方法:将被测试物与活细胞作用,正常兔血清作封闭,加入FITC—羊抗鼠IgG,固定液固定后荧光显微镜观察。结果:含抗T淋巴细胞白血病单抗腹水、单抗纯化物及单抗偶合物的效价分别为1∶10240、5 x 10-8mol/L和 5 x 10-8mol/L。结论:活细胞免疫荧光法是一种简单、快速、结果易观察的方法,可用于单抗偶合物制备过程中的抗体活性测定。Objective: To explore the value of the living cell immunofluorescence test in the study of immunoconjugate. Methods: At first, the tseted substances were added in the living cells. Then the normal rabbit serum was mixed with them as a blocking factor. After that, FITC-goat -anti -mouse IgG was added. Finally, the cells were mixed by the fixing solution and observed using the fluorescence micro- scope. Results: The titers of the ascetics, the purified monoclonal antibody and the immunoconjugate were 1:10240, 5 × 10-8 mol/L and 5 ×10-8 mol/L, respectively. Conclusions: The method is simple, rapid and the result is easy to observe. It is suitable for detecting the activities of monoclonal antibodies in preparing immunoconjugates.
