摘 要:目的:观察联合补充维生素C(VC)和维生素E(VE)对糖尿病大鼠高级糖化终产物(AGE)受体数量和结合力的影响。方法:采用链脲佐菌素腹腔注射SD大鼠建立糖尿病动物模型,对照组喂饲普通饲料,联合补充VC和VE组在纯营养素饲料配方中补充VC和VE,喂饲5w。用受体放射分析法,测定糖尿病大鼠腹腔巨噬细胞的AGE受体数和结合力常数。结果:联合补充VC和VE组大鼠腹腔每个巨噬细胞的AGE受体数是3.53×105,是糖尿病对照组的1.62倍;其结合力常数比对照组高出1倍,达到3.47×10-8M。结论:联合补充VC和VE可以上调AGE受体的功能,加快清除糖尿病大鼠体内的AGE,减缓糖尿病慢性并发症的发生及发展。Objective:To investigate the effects of vitamin C (VC) and vitamin E (VE) on number and affinity coefficient of receptors of advanced glycation end products ( AGE ) in experimental diabetic rats. Methods: SD rats were peritoneally injected with streptozotocin (STZ) to establish diabetic animal model. The number and affinity coefficient of AGE receptors in peritoneal macrophages of rats supplemented with VC and VE for 5w were investigated using radiolabeled ligand binding experiments. Results : Peritoneal macrophages of diabetic rats supplemented with VC and VE jointly displayed 3.53×105AGE receptors per cell, 1.62 times of control, and affinity coefficient Ka was 3.47×10-8 M, twice of control. Conclusion:The functions of AGE receptors in diabetic rats were significantly upregulated by VC and VE. AGE can be cleared more rapidly by supplementation with VC and VE jointly so that chronic complication can be prevented in diabetic rats.
