小麦新种质YW243抗条锈病基因的染色体定位

摘　　要：为鉴定小麦新种质YW243的条锈病抗性基因,用条中31号小种接种,对YW243与中国春的杂种F2群体进行了抗性遗传分析,并利用SSR分子标记技术对抗性基因进行了染色体定位。结果显示,YW243的条锈病抗性在与中国春的杂交组合F1、F2分离群体中呈一对显性基因的遗传模式;经过对264对微卫星引物的筛选,发现位于2BL上的两对引物Xgwm388和xgwm501能够在双亲和抗、感池之间扩增出特征带,并与抗性基因表现连锁,它们的遗传距离分别为27.9 cM和23.5 cM,说明抗病基因位于2BL染色体上。从系谱和抗谱分析,推测YW243的抗性基因不同于Yr5,可能是一个新的抗病基因或是Yr5的复等位基因。The losing of variety resistance to stripe rust (Puccinia striifarmis Westend f. sp. tritric) is an important factor causing periodical and massive stripe rust outbreak in wheat growing area. New strain of stripe rust pathogen has widely spread in varieties in China which lost resistance and therefore searching for new resistance resources is essential. Molecular marker technology could facilitate the identification and location of novel genes. The current study was undertaken using SSR technology in order to identify a stripe rust resistant gene in the novel resource YW243, and identify its chromosomal location. The F1 and F2 population derived from a cross between YW243 and cv. Chinese Spring were inoculated with stripe rust race Tiaozhong 31 at the seedling stage and the result indicated that the resistance was controlled by a dominant gene. After screening 264 pairs of wheat microsatellite primers, two markers Xgwm388 and Xgwm501 located on the 2BL linked to this gene with the genetic distance of 27. 9cM and 23. 5 was found, which indicated that the resistance gene was located on chromosome 2BL of wheat. Because YW243 was not related to Triticum spelta album in genealogy, the result also suggested that the resistance gene might be a new gene or the allele to Yr5 derived from Triticum spelta album.
