摘　　要：目的　探讨PC12细胞缺糖损伤时应激基因葡萄糖调节蛋白 (grp75 )的表达变化。方法　无葡萄糖培养基培养PC12细胞建立细胞缺糖损伤细胞模型 ,用RT -PCR、Westernblot和细胞免疫化学法分析缺糖损伤时 grp75的表达情况。结果　RT -PCR结果显示细胞缺糖损伤 3h起 grp75基因转录水平已显著增高 ,并在缺糖后 3～ 2 4h间持续上升 ;蛋白印迹法和细胞免疫化学法证实grp75在细胞缺糖过程中蛋白水平的表达明显高于对照组 ,且随着缺糖时间推延而进一步升高。结论　缺糖损伤时 ,细胞中的应激基因 grp75特异性上调表达。Objective: To explore the expression of grp75 in PC12 cell under glucose deprivation. Methods: PC12 cells were cultured with glucose free medium, and the expression of grp75 was determained by RT-PCR,Western blot and immunocytochemistry. Results: The expression of grp75 was upreglated since PC12 cells was glucose deprived for 3h,and the expression was increased markedly during the process of glucose derivation. Conclusion: grp75 was upreglated to generate cytoprotective effect under glucose deprivation.
