摘  要:目的采用国产的CD3单克隆抗体代替环胞菌素A,建立人淋巴母细胞株(LCL)。方法采静脉血用淋巴细胞分离液分离淋巴细胞,分别应用CD3单克隆抗体以及环胞菌素A作为免疫抑制剂,对比LCL建株情况。结果CD3单抗组经过4周的培养CD3+细胞由培养前的59.65%减低到52.78%,而环胞菌素A组升高至61.15%(P>0.05);对CD19+细胞的促进作用由培养前的5.39%提高到12.70%,而环胞菌素A为8.47%,两者的差异有显著性意义(P<0.01)。结论CD3单克隆抗体与环胞菌素A同样能够促进B淋巴样细胞株生成,防止体外B细胞的退化。Objective Using anti CD3 monoconal antibody to substitute cyclosporin A for the establishment of the human B lymphoblastoid cell line. Methods The lymphocyte were separated from the peripheral blood. Then the anti CD3 monoconal antibody and the cyclosporin A were used as immunosuppressive agent to eatablish human B lymphoblastoid cell line. Results After 4 week culture the percentage of CD3+cells in anti CD3 monoconal antibody group was decreased from 59 65%to 52 78%, but was increased to 61.15%(P >0.05)in cyclosporin A group. On the other hand, the percentages of CD19+cells in anti CD3 monoconal antibody group and cyclosporine A group were increased from 5.39%to 12.70%and 8.47%respectively. The difference between these two groups was significant(P< 0.01). Conclusion The anti CD3 monoconal antibody has the same enhancive effect as cyclosporin A on the establishment of B lymphocyte like cell line and can protect the B lymphocyte from regression in vitro.
