摘  要:目的在离体状态下观察川芎水煎剂和川芎挥发油对大鼠血流动力学的影响。方法应用Langendorff离体心脏灌流模型,在去除神经、体液因素影响的情况下,观察注射川芎水煎剂和川芎挥发油对大鼠心脏的影响。结果低剂量(02mg·mL-1)、中剂量(2mg·mL-1)组的川芎水煎剂在LVSP,LVEDP,HR,CF,+dp/dtmax,-dp/dtmax等各项血流动力学指标无明显改变,但中剂量(2mg·mL-1)组的水煎剂能显著地减少CF;高剂量川芎水煎剂(20mg·mL-1)可显著降低离体大鼠心脏的LVSP,+dp/dtmax,CF,HR,显著地升高-dp/dtmax;1,2,3μL挥发油对血流动力学各指标影响不明显。结论高浓度川芎水煎剂对离体大鼠心脏有负性变力和负性变时作用,并可使CF降低;川芎挥发油对大鼠心脏的血流动力学作用不明显。OBJECTIVE: To observe the hemodynamic effects of aqua decoction and volatile oil of Ligusticum chuanxiong Hort (LCH) on the isolated hearts of rats. METHODS: The actions of aqua decoction and volatile oil of LCH were investigated by using the Langendorff isolated rat heart perfusion model. RESULTS: With the concentrations of 0.2 and 2 mg&middotmL-1 of the decoction of LCH, the parameters of hemodynamics, left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP), heart rate (HR), coronary flow (CF), maximum rising rate of pressure (+ dp/dt max) and maximum declining rate of pressure (-dp/dtmax) did not show apparent changes. But the concentration of 2 mg&middotmL -1 of the decoction markedly decreased CF. With the concentration of 20 mg&middotmL-1 the decoction of herbal medicine of LCH, markedly decreased LVSP, + dp/dtmax, CF, HR and markedly increased-dp/dt max. Injection of volatile oil of LCH (1-3 μL), the parameters of hemodynamics was apparently changed. CONCLUSION: The results indicated that LCH decoction of high concentration has obvious negative inotropic action and can increase CF obviously; but the dynamic effect of LCH volatile oil was not clearly found.
