摘 要:目的 探讨镉对鲫鱼淋巴细胞DNA合成的影响及其卫生学意义。 方法 鲫鱼 72尾随机分成 12组 ,每组 6尾。设 2 .5、5、10、2 0和 40 μg/L五个氯化镉染毒组 ,水环境暴露 2 4h ,另设一个阴性对照组 ;在 10 μg/L暴露条件下 ,设 12、2 4、48、72和 96h五个时间效应组 ,另设一个 0时间对照组。各组在规定时间内心脏取血 0 .5ml,采用 3 H -胸腺嘧啶核苷 ( 3 H -TdR)掺入法测定鲫鱼 (Carassiusauratus)淋巴细胞中 3 H -TdR掺入量 (DPM值 )。 结果 各氯化镉染毒组的DPM值明显降低 ,与阴性对照组比较 ,差异有显著性 (P <0 .0 5 ,P <0 .0 1) ;另外 ,随着镉浓度的增高 ,各剂量效应组的DPM值逐渐下降 ,在实验浓度范围内 ,存在剂量 -效应关系 (P <0 .0 1)。DPM值与作用时间无相关性。 结论 镉可引起鲫鱼淋巴细胞DNA合成受抑 ,DNA合成可望作为水体污染生物检测指标之一。Objective To understand the effect of cadmium on DNA synthesis in carassius auratus lymphocytes and its implications of hygiene. Methods 72 carassiua auratua were randomly divided into 12 groups,6 carassius auratus each group. The carassius auratus were treated by being exposed in water for 24h with 2.5,5,10,20 and 40 μg/l cadmium chloride respectively,and set up a negative control group.At the concentration of 10 μg/l ,five time-effect groups(12,24,48,72 and 96 h) and a zero time control group were set up. 0.5 ml blood samples were obtained from heart in testing time and the value of 3H-TdR incorporation of carassius auratus lymphocytes was measured by using 3H-TdR incorporation. Results The value of DPM in all exposed groups decreased significantly compared to negative control group(P<0.05,P<0.01); In addition, the value of DPM of all exposed groups decreased gradually with dose of cadamium increasing and the dose-effect existed among tested concentrations(P<0.01). The value of DPM was irrelevant to exposure time. Conclusion Cadmium can inhibit DNA synthesis of Carassius auratus lymphocytes,it is expected that DNA synthesis might be one of bioassay indexes in water.
