摘  要:目的研究吡咯烷二硫代氨基甲酸盐(PDTC)对阿霉素(ADR)致大鼠心肌损伤的保护作用及作用机理。方法60只雄性Wistar大鼠,随机分为3组:ADR组、ADR + PDTC组和对照组。实验第30天,测定心肌组织总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性、超氧阴离子(O2·)及脂质过氧化物(LPO)水平,测定心肌组织中bcl-2、bax、p53 mRNA的表达。结果与ADR组比较,ADR + PDTC组心肌组织T-AOC升高、SOD和GSH-Px活性显著增加;心肌组织O2·及LPO水平显著降低;心肌组织中bcl-2 mRNA表达显著增加,bax、p53 mRNA表达显著降低。结论PDTC对ADR致大鼠心肌损伤有保护作用,其机制可能与增加抗氧化酶的活性,减少活性氧的产生,进而调节凋亡相关基因的表达有关。Objective To study the protective effects of pyrrolidine dithiocarbamate(PDTC) on cardiomyopathy induced by adriamycin(ADR) in rats and its mechanisms. Methods Sixty male Wistar rats were randomly divided into three groups: ADR, ADR+PDTC and control groups. After 30-day experiment, myocardium activity of T-AOC, SOD and GSH-Px and amount of superoxide anion and LPO were examined; myocardium bcl-2, bax, p53 gene expression were examined. Results Compared with ADR group, myocardium activity of T-AOC,SOD and GSH-Px increased significantly; superoxide anion and LPO in myocardium decreased significantly in ADR+PDTC group. Moreover upregulation of bcl-2 and downregulation of bax and p53 were observed in ADR+PDTC group. Conclusions PDTC has a preventive effect on cardiomyopathy induced by ADR, which is probably associated with improving activity of antioxidant enzymes, decreasing reactive oxygen species and regulating apoptosis gene expression.
