摘  要:目的探讨血管内皮生长因子(VEGF)在口腔鳞状细胞癌侵袭转移中的作用。方法采用3H~TdR 掺入粘附试验测定VEGF对体外培养的口腔鳞状细胞癌细胞系TSCCa细胞同质性粘附作用以及Boyde Chamber 观察VEGF诱导口腔鳞状细胞癌细胞转移作用。结果1ng/ml、5ng/mlVEGF诱导TSCCa细胞60min、90min、120min,3H-TdR掺入实验(dpm/min)分别为1773.67±289.64、2180.32±326.321、2256.43±310.31和1687.38±226.24、2045.68±273.26、1891.52±213.84,10ng/ml VEGF诱导TSCCa细胞60min、90min、120min,3H-TdR掺入实验(dpm/min)分别为1436.69±326.03、1380.32±201.04、1228.56±237.07,显著低于对照组60、90、120min的2184.49±314.72、2746.53±255.17、3560.14±377.35(P<0.05或0.01),用VEGF 1ng/ml、5ng/ml、10ng/ml培养口腔鳞状细胞癌2h,Boyde Chamber小室下室浸润的口腔鳞状细胞癌细胞数分别为6.67±1.78、17.17±2.38、22.33±2.54×104/ml,分别高于对照组2.48±1.02×104/ml(P<0.05或0.01)。结论VEGF可以促进口腔鳞状细胞癌细胞转移,与VEGF降低口腔鳞状细胞癌细胞的同质性粘附作用有关。Objective To determine the role of vascular endothelial growth factor (VEGF) on the invasive metastasis of oral squamous cell carcinoma cells. Methods The effects of VEGF on homotypic adhesion and metastasis in oral squamous cell carcinoma cells were examined with 3H- TdR infiltration and Boyder chamber. Results After 60,90, 120 minutes induction by 1ng/ml VEGF, the values (dpm/min) of 3H-TdR infiltration in TSCCa cells were 1773.67± 289.64,2180.32±326.321, and 2256.43±310.31, respectively. The values for 5ng/ml VEGF induction were 1687.38±226.24,2045. 68±273.26, and 1891.52±213. 84. The values for 10ng/ml VEGF induction were 1436. 69 ±326.03,1380. 32±201.04 and 1228. 56±237. 07. The 10ng/ml group showed significantly lower values than that in control group ( P < 0.01). The cells in lower chamber were (6. 67±1.78)×104/ml and (22.33± 2.54 )×104/ml after cultured for 2 hours with Ing/ml,5ng/ml and 10ng/ml of VEGF. Those values were remarkablly higher than the values for the control group of (2. 48±1.02)×104/ml ( P < 0.01) . Conclusion VEGF can decrease homotypic adhesion of oral squamoua cell carcinoma cells and may be a cause of the metastasis.
