摘  要:目的分离培养骨髓间充质干细胞(mesenchymalstemcells,MSCs)并进行鉴定。方法取周龄大鼠骨髓用percoll分离液分离,DMEM(含20%胎牛血清)培养、传代,碱性磷酸酶和苏丹Ⅳ染色,流式细胞仪测MSCs表面标志CD44和CD45。结果碱性磷酸酶染色和苏丹Ⅳ染色阴性,CD44、CD45表达甚微。结论所分离培养的细胞为未分化的具有较强增殖能力的细胞,可认为是间充质干细胞。Objective Bone marrow mesenchymal stem cells(MSCs) were separated, cultured and identified. Methods bone marrow of a-week-old rat was obtained, separated with Percoll, then cultured in DNEM medium containing 20% FCS and passaged. Then surface markers of MSCs, CD44 and CD45 were detected with Flow cytometry analysis and Sudan IV and AKP stain were performed. Results CD44、CD45 expressed lightly, and the results of Sudan IV and APK stain were negative. Conclusion The cultured cells have strong ability of proliferation with undifferented characteristic, which may be considered as MSCs.
