摘 要:目的 研究雷帕霉素 (RPM )和他汀类药物在大鼠移植心脏慢性血管病变中的作用。方法 建立大鼠心脏移植慢性排斥 (CR )模型 ,并分别给予赋形剂、RPM、他汀类药物及环孢素(CsA)等药物处理 ,应用组织学方法观察不同药物对于移植物慢性血管病变的作用。结果 与对照组相比 ,RPM处理组的移植物慢性血管病变程度显著减轻 (P <0 .0 1) ,CsA处理组有所减轻(P <0 .0 1) ,而他汀类药物组动物未见明显减轻。结论 RPM可以明显抑制血管平滑肌细胞增生 ,预防新内膜形成和移植物慢性血管病变 ;本实验未能证实他汀类药物的抑制作用。Objective To study the effects of Rapamycin and Statin on chronic transplant vasculopathy in rat cardiac allografts.Methods After the establishment of chonic rejection model in rat,the vehicle (control group),Rapamycin,Statin analogs-STA352 and CsA were administered respectively in different groups.The effects of different drugs on the chronic transplant vasculopathy were observed and analyzed by histological methods.Results As compared with animals in control group,graft vascular remodelling of vasculopathy was significantly inhibited in Rapamycin treated rats (P< 0.01).In CsA treated animals the vascular remodeling was also significantly inhibited (P <0.01).However there was no significant difference in vasculopathy between STA352 treated animals and control animals.Conclusion Rapamycin can inhibit proliferation of vascular smooth muscle cells and prevent neointimal formation and transplant arteriosclerosis in this model.The present data do not provide strong evidence for inhibition effect by STA352.
