摘 要:目的:探讨抽吸负压对所抽吸脂肪颗粒活性的影响,明确脂肪移植术中是否可用吸脂机获取脂肪。方法:用 60ml 注射器(26~40kPa)和吸脂机(70~80kPa)连接相同管径不同侧孔的吸管抽取两组脂肪颗粒(Ⅰ、Ⅱ组),同时进行葡萄糖转移实验:即每组取 12 个标本,5ml/ 标本,置于培养皿中,加入 10ml 含糖 DMEM,孵育 1h 后,检测 DMEM 中葡萄糖浓度的变化,以此代表脂肪颗粒葡萄糖转移量即活性,t 检验比较两组活性有无差异。两组样本各取 10ml 送病理学检查,比较两组切片完整细胞数有无差异。用 60ml 注射器和吸脂机(压力同上)连接相同侧孔不同管径的吸管抽取两组脂肪颗粒(Ⅲ、Ⅳ组),同时进行葡萄糖转移实验和病理学检查,方法同前。取Ⅰ、Ⅱ组脂肪颗粒各 5ml 分别注射于两组裸鼠(16 只 / 组)颅顶部皮下,每只注射 0.3ml,15 周后取出,比较二组移植物残留体积和重量及组织学特性有无差异。结果:60ml 注射器(26~40kPa)和吸脂机(70~80kPa)连接相同管径不同侧孔吸管抽取的脂肪颗粒葡萄糖转移量差异显著,前者小于后者(P<0.05),病理切片完整细胞数差异显著,前者大于后者(P<0.05);而连接相同侧孔不同管径吸管抽取的脂肪颗粒葡萄糖转移功能及病理切片完整细胞数均差异不显著(P>0.05)。植入裸鼠颅顶部皮下的Ⅰ、Ⅱ组样本,15 周后。Objective To examine the effect of suction pressure on aspirated fat viability to determine if machine suction can be used to harvest fat for grafting. Methods We harvested two groups (ⅠandⅡ) of fat with machine (usually 70~80kPa) and 60ml syringe (26~40kPa) jointing same diameter but different distal opening cannulas, made them suffered glucose transportation test which was as follows putting two groups of fats into DMEM containing glucose to incubate for 1 hour and then detecting the change of glucose consistence in DMEM which represented the glucose transportation quantity,in other words, the viability of fat. The results were analyzed and compared with t test. The two groups of fat were put in histopathologic examination and the amounts of intact adipocytes on two groups of slides were compared with T test. We transplanted the two groups of fats subcutaneously on the nude mice skull (16 mice/ group, 0.3ml fat/ mouse) and detect the difference of weight and volume retention and histological characteristic between two groups after 15 weeks. We also harvested other two groups (Ⅲ and Ⅳ) of fat with machine (usually 70~80kPa) and 60ml syringe (26~40kPa)jointing same distal opening but different diameter cannulas and put them in glucose transportation test and histopathologic examination ditto. The results were analyzed and compared with t test. Results The glucose transportation quantities were significantly different between groupⅠand groupⅡ, machine more than syringe, (P<0.05); the intact cells on pathologic slides were also significantly different between groupⅠand groupⅡ, but syringe more than machine, (P<0.05) . While the glucose transportation quantity and intact cells on histopathologic slides were all same between group Ⅲ and group Ⅳ(P>0.05). The histological characteristic and weight and volume retention of groupⅠand groupⅡ of fat grafts are all same (P>0.05). Conclusion The effect of machine suction pressure (usually 70~80Kpa) on fat viability is same as that of syringe
