摘 要:目的:初步探索多种生长因子、DMEM 培养液对植入脂肪成活率的影响,为临床应用提供参考。方法:将同条件获取的脂肪颗粒 40ml 纯化后,等量分 5 组,分别于生理盐水(对照组)、DMEM 液、含 20ng/ml bFGF 的生理盐水、含 20ng/ml IGF-1 的生理盐水、含 20ng/ml VEGF 的生理盐水(以上四组为实验组)中浸润 1h。浸润后去杂质,注射于裸鼠背部皮下,每组注射 5 只裸鼠,每只注射 6 点,每点 0.2ml,15 周后,取出移植物,测残留体积和重量,并送病理学检查。方差分析检验比较各实验组与对照组移植物残留体积和重量的差异。结果:DMEM 组的移植物残留体积和重量明显较对照组多(P<0.05),其他实验组移植物残留体积和重量与对照组无明显差异(P>0.05),各组移植物组织病理学特性无明显差异。结论:DMEM 可促进植入脂肪的组织量维持,bFGF、IGF-1、VEGF 等生长因子在本实验条件下显示出促进植入脂肪成活的作用。Objective We research the effects of DMEM,bFGF,IGF-1 and VEGF on the grafted fat survival to offer reference to clinical practice. Methods We divided fat harvested under same condition into 5 groups and infiltrated the 5groups of fat respectively in saline, DMEM, saline containing 20ng/ml bFGF, saline containing 20ng/ml IGF-1, saline containing 20ng/ml VEGF for one hour. And then we transplanted 5 groups of fat subcutaneously on the back of 5 groups of mice, 5 mice per group, injecting 5 points per mouse, 0.2ml per point. After 15 weeks, we dissected fat grafts, measured their volume and weight retention and put them in histopathologic examination. We compared volume and weight retention of each experimental group with that of control group. Results The volume and weight retention of the group DMEM was significantly more than that of control group(P<0.05). The volume and weight retention of the other experimental groups were all same as that of control group(P>0.05). The histopathologic characteristics of all grafts were same. Conclusion DMEM can improve the volume and weight maintenance of graft. bFGF,IGF-1 and VEGF didn′t display the effect of improving fat survival in our experiment condition.
