当归补血汤对高眼压兔视网膜神经节细胞凋亡的抑制作用

摘　　要：目的在眼压得到一定程度控制情况下观察当归补血汤对实验性高眼压兔视网膜神经节细胞凋亡的抑制作用。方法用04%的复方卡波姆前房注射造成兔实验性高眼压模型。39只兔分入正常组、Timolol组(T)、Timolol加中药组(TC)和高眼压未治疗组(EP)。观察4、6、8周后处死,制作病理切片,通过计数凋亡视网膜神经节细胞数和其超微结构变化评估当归补血汤对凋亡的抑制作用。结果MG-P-MY标记的切片凋亡视网膜神经节细胞数TC组为067±082(个/张),明显低于T组4±212(个/张)和EP组529±304(个/张,P<001);TUNEL-N末端标记凋亡的视网膜神经节细胞数数正常组和给药组之间差异无显著性,但明显低于EP组(P<001);视网膜神经节细胞的超微结构TC组细胞变性程度明显轻于T组和EP组,仅见部分细胞器轻度扩张,未见凋亡的典型改变。结论在眼压得到一定程度控制情况下当归补血汤对实验性高眼压所致的视网膜神经节细胞的凋亡具有抑制作用。OBJECTIVE To investigate the anti-apoptotic function of dangguibuxuetang(当归补血汤)on the retinal ganglion cells(RGCs)of rabbits with experimental ocular hypertension controlled to some degree.METHODS Experimental ocular hypertension wa produced by injecting 0.4% carbomer into anterior chambers.39 rabbits were divided into normal group,timolol group(T),timolol plus dangguibuxuetang group(TC),and untreated control group(EP).To evaluate the results by the number of apoptotic RGCs and its ultrastructure they were killed respectively at 4,6,and 8 weeks.The retinas were obtained for pathological analysis.RESULTS MG-P-MY stained apoptotic RGCs:TC group 0.67±0.82 positive cells/section,the number of cells was lower than EP group 5.29±3.04/section (P<0.01).T group 4±2.12/section.There was not significant difference between T group and EP group,but higher than TC group (P<0.01).TUNEL-labeled RGCs:there were no significant difference within normal and two treated groups,but they all were significant lower apoptotic RGCs than EP group (P<0.01).RGCs ultrastructure:the degeneration and vacuolization of RGCs in TC group was slighter than T and EP groups,no remarkable apoptotic cells had been seen in TC group.CONCLUSION With IOP controlled to some degree,dangguibuxuetang can protect RGCs and has the function of anti-apoptosis.
