摘 要:目的通过去势大鼠骨质疏松骨折模型,研究骨质疏松骨痂软骨细胞的增殖及IGF-I的表达情况。方法建立去势大鼠骨质疏松及骨折模型,在术后10~40d内取材,行IGF-I和PCNA免疫组化染色,观察骨质疏松骨折骨痂内软骨细胞的增殖及IGF-I的表达情况。结果骨质疏松性骨痂组织中IGF-I早期上调表达,可以促进骨膜生发层细胞和骨折断端间充质细胞增殖、分化;IGF-I在骨折后期一部分成熟软骨细胞中表达,可能延缓了软骨细胞向终末分化,在维持软骨细胞表型过程中发挥一定的作用。结论IGF-I在骨质疏松性骨折愈合过程中可能发挥着重要的调节作用。Objective: To study the proliferation of chondrocyte and the expression of insulin-like growth factor I (IGF-I) in the osteoporotic callus by means of ovariectomied rat fracture experimental model.Method: An osteoporotic model by ovariectomied rat was made, and then the right femur middle was fractured following up with the medullary pin fixation.The rats were sacrified from 10~th day to 40~th day after femur fracture.The change of chondrocyte’ proliferation and expression of IGF-I in the osteoporotic fracture callus was observed by immunohistochemistry staining of PCNA and IGF-I.Result: IGF-I up-regulating in the early stage of osteoporotic fracture callus facilitated mesenchymal cell surrounding the fracture and in the greminative layer of periosteum to proliferate and differentiate.It suggested that IGF-I might be delayed the chondrocyte further differentiation and may have an ability to maintain the phenotype of chondrocyte by expressing IGF-I in some of maturated chondrocytes.Conclusion: IGF-I may play an important regulating role in osteoporotic fracture healing.
