摘 要: 目的 建立并探讨媒染剂依来铬天青B测定铁的方法及方法学评价。方法 在pH值4.75条件下, 铁离子、依来铬天青B与阳离子活性剂形成蓝色的三元络合物,其吸光度与血清铁含量呈正比。结果 线性范 围达89.500μmol/L,批内与批间变异系数(CV)分别为1.54%和3.03%,回收率为97.30%和102.57%,铜、锌 各50μmol/L,铅、镉各10μmol/L及胆红素100μmol/L时均不干扰反应,本法(Y)与亚铁嗪法(X)比较相关性 好,Y=1.021X-1.42,r=0.993。试剂稳定,在37°C下放置10d后,对结果无影响。结论 本法为单一试剂,可 用于自动分析或手工操作,抗干扰性能好,灵敏度高,各项方法学评价指标均能满足常规测定血清铁的要求。Objective To establish and evaluate a method of serum iron determination using a mordant dye eriochrome azurol B. Methods Iron reacts with eriochrome azurol B and cationic detergent to from a blue ternary complex, which was directly proportional to the iron content of the sample. Results The linearity was up to 89.500 μmol/L. The within-run CV and between-run CV were 1.54% and 3.03% respectively. Recovery rates were 97.30% and 102.57%. Cu 2+, Zn 2+ up to 50 μmol/L, Pb 2+, Cd 2+ up to 10 μmol/L and bilirubin up to 100 μmol/L showed no interference. Correlation between results obtained by this method (Y) and ferrozine method (X) was Y=1.021X-1.42, r=0.993. Conclusions The method is simple, sensitive and stable. It is suitable for the routine manual or automatic analysis of iron in serum.
