流式细胞术及图像分析术测定TNF_a对Eca—109细胞DNA含量的影响

摘　　要：作者用重组人TNF.(rh—TNF.)及其与化疗药物合用对人食管鳞状上皮癌细胞系(Eca—109)进行了体内外细胞毒性试验,对体外培养Eva—109细胞进行流式细胞术及自动图像分析术测定。结果表明,TNF.主要作用于Eca—109细胞周期的G_0/G_1期,阻滞细胞向S期转化,抑制DNA合成。随给药时间的延长,实验组同对照组相比,各周期细胞相对量(即DNA含量)百分率有显著差异(P<0.55)。并提示TNF.与某些抑制DNA合成或周期特异性药物合用可增强各自的作用。本文结果也表明,两种测量术细胞增殖周期的改变和DNA含量变化的直方图基本吻合。Flow cytometry and image analyses were performed to investigate the cell cycle distribution and DNA content of Eca—109 cells in vitro, Which were treated by TNF_α and/or 5—Fu or PYM. The results showed that TNF_α acted mainly on the phase G_0/G_1 in cell cycle and blocked progression into S stage, and DNA synthesis was inhibited, especially when combined with the anticancer drugs. This observation indicated that TNF_α combined with DNA systesis inhibitor such as 5—Fu may enhance the anti—tumor effect.
