植物Brazzein基因的合成及其克隆

摘　　要：根据从西非植物PentadiplandrabrazzeanaBaillion果实中分离的甜蛋白Brazzein(Bra)的成熟区氨基酸序列,按照酵母密码子偏好人工合成了4对末端具粘合位点的寡聚核苷酸序列,经连接、PCR扩增后得到了179bp的Brazzein类似物的编码序列,插入到pPIC9K载体构建成重组表达载体pPIC9K Bra.According to the amino acid sequence of Brazzein isolated from the fruit of Pentadiplandra brazzeana Baillion,four pairs of oligoneucleotide with cosmic site were synthesized using yeasty biased codons. After linkage and PCR, the 179 bp coding region of des-pGLU1-Brazzein was obtained and inserted into pPIC9K which results in the recombinant expression vector pPIC9K-Bra.
