摘 要:本试验采用纤维滤袋技术,应用胃蛋白酶、胰蛋白复合酶、纤维素酶、猪小肠冻干粉和牛瘤胃液多酶体系进行体外养分消化试验。测定玉米蛋白粉的粗蛋白质、干物质和能量消化率,并与相应的猪体内养分表观消化率进行比较。结果表明三步酶法(胃蛋白酶+胰蛋白复合酶+牛瘤胃液或胃蛋白酶+胰蛋白复合酶+纤维素酶)技术能较准确模拟体内全消化道养分消化。体外养分消化率受样品的物理化学特性、样品的粒度、酶活性、酶浓度、底物浓度、pH、温度、动态时间及体外技术等综合因素影响。体外和体内粗蛋白质消化率相关系数为0.72,总能消化率相关系数为0.67。The filter bag technique in vitro was used to determine the digestibilities of gross energy,dry matter and crude protein of corn gluten meal.Pepsin,pancreatin,intestinal fluid,rumen fluid and cellulase were used in incubation.The results indicated that filter bag technique was much more convenient and fast.The digestion in the three - step multi - enzyme systems(pepsin + pancreatin + rumen fluid or pepsin + pancreatin + cellulase)was more similar with in vivo.Furthermore,In vitro,the nutrient digestibility was influenced by some factors such as feedstuffs characteristics,sample particle,enzyme activity,enzyme concentration,stirring frequency,stirring time,incubating temperature and buffer solution pH.The correlation coefficient of the crude protein digestibility and the gross energy digestibility was 0.72 and 0.67 between in vivo and in vitro respectively.
