摘 要:目的 研究低温冻结和液态冷藏条件下.海藻糖对血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、γ-谷氨酰转肽酶(γ-GT)、碱性磷酸酶(ALP)的活性保护作用,并探讨海藻糖对酶的最佳保活浓度,以提供一种多酶质控品的良好稳定剂。方法 采用酶动力学方法分别在30 d、60d、120d、240d、360d测定四种酶活力,并计算酶的保活率(%)。结果在360d时,海藻糖保护组的四种血清酶的平均保活率为93.7%.而无海藻糖保护组的四种血清酶的平均保活率为67.7%,海藻糖组的保活率是无海藻糖组的1.38倍,尤其是海藻糖对ALT的保活作用尤为显著,其保活率是对照组的2.55倍。海藻糖对酶的保活最佳浓度为(0.10~0.25)mol/L。,低于 0.10 mol/L。或高于 0.30 mol/L.则保活效果不佳。对液态下海藻糖的保活效果进行时效观察,结果对ALT有较明显的保护作用,保活率是其对照组的2.06佰。结论 海藻糖对酶在冻结状态下具有保活作用,是酶蛋白良好的稳定剂。Objective To study the effect of trehalose protection for four kinds of serum enzymes at freezing and refrigeration, and to explore the range of concentration of trehalosc, under which the best activity concentration of enzymes can be maintained ,so as to develop a good stabilizer of multienzymes QC material. Methods The activities of four kinds of enzymes were measured with kinetic at 30 d.60 d.120 d.240 、d360d respectively, and protection activity rate(%) of enzymes was calculated. Results At 360 d,the average PAR of trehalose group was 93. 7%,and of non-trehalose group was 67. 7%. The PAR of the trehalosc group is 1. 38 times as large as non-trehalose group. Especiaiiy.ALT activity was extraordinary remarkable under trvhalose protection. And the PAR of trehalose group is 2. 55 times as large as non-trehalose group. The best activity concentration of enzymes was between 0. 10 mol/L. and 0. 25 mol/L. Conclusion The trehalose is an important protective reagent to the activity of enzymes and also a good stabilizer of enzymes protein.
