摘 要:目的 建立DNA指纹技术对近交系大鼠遗传监测的方法。方法 采用DNA指纹图法对国内已知的7个品系 9个近交系大鼠群体进行了分析 ,并对相同DNA进行了多次DNA指纹图重复实验。结果 (1)不同品系之间DNA指纹图差异较大 ,其平均图带数为 16 .36 0± 2 .178,共有带率为 0 .0 6 1± 0 .0 0 8,相似系数为 0 .0 6 2± 0 .0 0 8,相同DNA指纹图概率为 3.6 91× 10 - 2 3 。 (2 )同一群体不同个体之间DNA指纹图带的相似系数和共有带率 (除SHR(哈 )和WKY(哈 )小于 0 .7外 ) ,其他均在大于 0 .9。 (3)不同地区同一SHR和同一WKY品系间DNA指纹图也存在不同 ,其相似系数和共有带率均小于 0 .6。 (4 )相同DNA不同次制作的DNA指纹图谱基本一致 (P >0 .0 5 )。Objective In order to set up the method of DNA fingerprints in the heredity monitoring of inbred rats. Methods\ National nine groups of seven strains were analysed using DNA fingerprints, and the same DNA were repeatedly tested by DNA fingerprints. Results\ (1)There were great diversity of DNA fingerprints in various strains, whose average bands were 16 360±2.178,band sharing level was 0.061±0.008,and the similarity coefficient was 0.062±0.008,the probability of an identical DNA fingerprints in them was 3.691×10 -23 . (2) Except that the similarity coefficient and band sharing level of SHR(Haerbin)and WKY(Haerbin)were less than 0.7 in various individuals of the same group, the remainder were more than 0.9. (3) It was diverse between the same inbred SHR strains or WKY strains in different areas, the similarity coefficient and band sharing level being less than 0.6. (4) DNA fingerprints from the same DNA were almost consistent in repeated experiments ( P >0 05). Conclusions\ Feasibility of DNA fingerprints in the heredity monitoring of inbred strain rat was proved.
