摘  要:本试验探讨了重组人白细胞介素-1β(rhIL-1β)对小鼠桑椹胚玻璃化冷冻后体内外发育的影响。结果表明:当rhIL-1β添加浓度为5、50、500IU/ml时,对解冻后的胚胎经体外培养,发育率(87%~100%)与对照组(94%)相比无显著性差异(P>005)。而浓度提高到1000IU/ml时,胚胎的发育受到抑制。解冻后胚胎在含有5、50、500IU/ml浓度的rhIL-1β培养液中培养3~4h后移植于假妊娠3d的受体鼠,移植妊娠率(86%、100%、83%)高于对照组(56%),特别是50IU/ml处理组与对照组有极显著差异(P<001)。而浓度为1000IU/ml处理组的胚胎移植妊娠率(30%)低于对照组。试验证明,添加浓度为5~500IU/mlrhIL-1β的培养液,对解冻后胚胎的体外发育率无影响,而且能够显著提高移植妊娠率。The present study examined effects of rhIL-1 β on the development of mouse froze n-thawed morulae. The frozen-thawed embryos were cultured in mKRB supplemented w ith 5IU/ml, 50IU/ml and 500IU/ml rhIL-1β, the rates of development were 87%, 97 % and 100% respectively. In comparisons between the presence or absence of rhIL- 1β, no differences were observed in the development rates. Further increase fro m 500IU/ml to 1000IU/ml rhIL-1β reduced the development rate. The frozen-thawed morulae were cultured in mKRB supplemented with 5IU/ml, 50IU/ml and 500IU/ml fo r 3~4h, then transferred into 3-day pseudopregnant recipients. The pregnant rat es in three groups were significantly higher than that in control. However, whe n the concentration of rhIL-1β increased to 1000IU/ml, the pregnant rate was si gnificantly lower than that in control. The results show that the presence of rhIL-1β in mKRB with the concentration from 5IU/ml to 500IU/ml had no effect on the devel opment of mouse frozen-thawed morulae in vitro, but it improved the pregnant rat es after the embryos were transferred into recipients.
