摘 要:目的:探讨基质金属蛋白酶9(matrixmetalloproteinase-9,MMP-9)、转化生长因子β1(TGF-β1)及其受体TβR-Ⅰ在人冠状动脉粥样斑块中的表达水平,以及三者与粥样斑块稳定性的关系。方法:收集人冠状动脉冠状动脉斑块组织共41例,常规石蜡包埋切片,作苏木精-伊红染色,根据镜下结构将斑块组织分为稳定组和不稳定组,采用免疫组化方法分析MMP-9,TGF-β1,TβR-Ⅰ在人冠状动脉粥样斑块中的表达,并通过计算机图像分析系统进行定量。结果:不稳定斑块内,MMP-9的表达要明显强于稳定性斑块,TGF-β1则明显弱于稳定性斑块,两者在斑块内的表达呈明显的负相关(面密度,r=-0.332,P=0.034;吸光度,r=-0.373,P=0.016),TβR-Ⅰ的表达无明显差异。结论:MMP-9,TGF-β1同斑块的稳定性密切相关,MMP-9是形成不稳定斑块的重要的促进因素,TGF-β1则是重要的斑块稳定因素。AIM:To investigate the expression levels of matrix metalloproteinase-9(MMP-9),transforming growth factor-beta 1(TGF-β1) and its receptorⅠ(TβR-Ⅰ) in human atherosclerotic plaque and their relationship with the stability of atherosclerotic plaque. METHODS:The specimen of human coronary artery atherosclerotic plaque were obtained from 41 patients undergoing coronary endarterectomy,and were routinely paraffin embedded and sectioned,then stained with hematoxylin and eosin(HE).They were divided into stable plaque group and unstable plaque group according to the structure under light microscope,expressions of MMP-9,TGF-β1 and TβR-Ⅰin the human atherosclerotic plaque were analyzed with immunohistochemical staining,and then quantitative analysis was performed with computer image analyzing system. RESULTS:The expression of MMP-9 in the unstable plaques was much stronger than in the stable ones,but the expression of TGF-β1 was obviously weaker in the unstable plaques than in the stable ones.There was a negative correlation between the expression of MMP-9 and TGF-β1 in plaques(areal density:r=-0.332,P=0.034;absorbance:r=-0.373,P=0.016),and no obvious difference was observed in the expression of TβR-Ⅰ. CONCLUSION:MMP-9 and TGF-β1 are closely related with the stability of plaques.Enhanced production of MMP-9 may participate in the formation of unstable plaque,while TGF-β1 maybe an important stabilizing factor for stability of plaques.
